摘要
利用反相高效液相色谱洗脱,高丝氨酸作为内标,邻苯二甲醛柱前衍生和荧光检测器检测南方鲇延髓面叶4种氨基酸类神经递质天冬氨酸(Asp)、谷氨酸(Glu)、甘氨酸(Gly)和γ-氨基丁酸(GABA)的含量.结果表明,4种氨基酸在40 min内洗脱完全,分离效果良好;氨基酸浓度与峰面积比值呈良好的线性关系,其相关系数不低于0.999 2.该方法简便、特异性强、重复性好,适用于南方鲇延髓面叶氨基酸类神经递质的含量测定.
HPLC system with fluorescence detection (FL) was used to examine amino acid derivatives, including aspartate, glutamate, glycine and γ-aminobutyric acid in facial lobe of Silurus meridionalis Chen formed by a reaction with o-phthalaldehyde (OPA) in the presence of the internal standard homose-rine. The result showed that precise separation of the four amino acids was achieved using isocratic elution within 40 min. Amino acid concentration and peak area ratio was a good linear relationship with correlation coefficients not less than 0. 999 2. Therefore, the method is simple, sensitive and has strong specificity and good repetition. It is suitable for the content determination of amino acid neurotransmitters in facial lobe of Silurus meridionalis Chen by HPLC-FL.
