摘要
[目的]根据白酒丢糟原料的特点,选取优势菌种构建丢糟多菌发酵复合处理体系,探讨各菌种液态培养最优条件。[方法]基于前期白酒丟糟发酵试验结果,产朊假丝酵母、绿色木霉和黑曲霉为丟糟复合微生物菌剂的优选菌种。经单因素试验和正交试验确定了产朊假丝酵母、绿色木霉和黑曲霉菌的最佳液态培养条件。[结果]以7°Bx麦芽汁为基础培养基,各菌最佳培养条件为:对于产朊假丝酵母,添加1.5%的硫酸铵、接种量为5%、培养时间为16h;对于黑曲霉,添加0.3%的氨水、接种量为15%、培养时间为18h;对于绿色木霉,添加0.5%的氨水、接种量为15%、培养时间为22h。[结论]在该培养条件下各单菌发酵液的活菌数量可达到1010个/ml以上,经喷雾干燥制备的微生物菌剂的活菌数量可达到109个/g以上。
[Objective]According to the character of wine waste lees,the dominant strains were selected to establish the system of mixed microbial fermentation.Besides,the optimal liquid-state culturing conditions were studied.[Method]Based on the results of the previous research,Candida tropicalis,Trichoderma viride and Aspergillus niger were the excellent strains of waste lees by mixed microbial agents.The optimal liquid-state culturing conditions were defined by single factor test and orthogonal test.[Result]The optimal culturing conditions of the strains were as following:the basic medium contains 7°Bx malt extract.For Candida utilis,amount of ammonium sulfate was 1.5%,inoculum size was 5%,and fermentation time was 16 h.For Aspergillus niger,amount of ammonia was 0.3%,inoculum size was 15%,and fermentation time was 18 h.For Trichoderma viride,amount of ammonia was 0.5%,inoculum size was 15%,and fermentation time was 22 h.[Conclusion]Under these culturing conditions,the living bacteria number in the fermentation liquid of the singal strains reached more than 1010 viable cells per ml.And the living bacteria number of the microbial inoculants after prepared by spray drying was up to 109 viable cells per g.
