摘要
[目的]优化玉米须多糖sevag法脱蛋白质的工艺条件。[方法]在单因素试验基础上,通过4因素3水平正交试验研究脱蛋白质工艺。4因素为正丁醇与氯仿体积比、多糖样液与sevag试剂体积比、振荡时间和脱蛋白次数。[结果]最优脱蛋白工艺为氯仿与正丁醇比5∶1,样液与试剂比2∶1,振荡时间8min,脱蛋白次数4次。[结论]在最佳脱蛋白条件下,蛋白脱除率为64.2%,多糖损失率为34.3%,多糖纯度由11.5%提高为32.3%。
[Objective] The research aimed to optimize the deproteinization process conditions of polysaccharides of corn stigma by sevag method.[Method]On the basis of single factor experiment,the deproteinization process was tested by orthogonal experiment of 4 factors and 3 levels.4 factors were volume proportion of n-butanol and chloroform,volume ratio of polysaccharide liquid and sevag reagent,shaking time and times of deproteinization.[Result]The optimal process of deproteinization was as follows:the volume proportion of n-butanol and chloroform was 5:1,sample liquid and sevag reagent ratio was 2∶ 1 and shaking time was 8 min,deproteinization times was 4 times.[Conclusion] Under the best conditions,the rate of deproteinization was 64.2% and the loss of polysaccharide was 34.3% .Polysaccharide purity increased from 11.5% to 32.3% .
