摘要
[目的]建立曼地亚红豆杉根中紫杉醇含量的HPLC测定方法,揭示不同年龄曼地亚红豆杉根中紫杉醇含量的动态变化。[方法]采用高效液相色谱法,Phenomenex Luna C18色谱柱(250.0mm×4.6mm,5μm),流动相为甲醇-乙腈-水,流速为1.0ml/min,检测波长为227nm,柱温为40℃,进样量为20μl。[结果]紫杉醇在此色谱条件下分离良好,其质量浓度与峰面积在1.00~32.00mg/L(r=0.9999)呈良好的线性关系,平均加样回收率为100.3%,RSD值为1.6%。不同年龄曼地亚红豆杉根中紫杉醇含量差异较大。[结论]首次建立了高效液相色谱法测定曼地亚红豆杉根中紫杉醇含量的方法,该方法准确可靠,可用于其根中紫杉醇的含量检测。
[Objective]To develop a reliable HPLC method for determining the dynamic change of taxol content in root of Taxus media in different ages.[Method]The HPLC conditions were as follows:phenomenex Luna C18 column (250.0 mm×4.6 mm,5μm) ,eluent of methanol-acetonitrile-water,flow rate of 1.0 ml/min,detection wavelength of 227 nm,column temperature of 40 ℃ and injecting amount of 10 ul.[Result]The taxol was well separated under this condition.In the range of 1.00-32.00 mg/L (r=0.999 9) ,the linear relationship was good,and the average recovery rate and RSD value were 100.3% and 1.6% respectively.The content of taxol in root of Taxus media in different ages varied.[Conclusion]A reliable and convenient method was firstly established to determine the taxol content in root of Taxus media.
