清营解表合剂对流感病毒A/PR/8/H1N1感染小鼠肺组织炎性细胞因子IL-6,IL-10,TNF-β动态表达的影响 被引量:1

摘要 目的动态观察清营解表合剂对流感病毒感染小鼠Th类细胞因子表达的影响,探讨该方抗流感病毒感染的免疫机制。方法以流感病毒亚洲甲型鼠肺适应株(A/PR/8/H1N1)感染小鼠为模型,采用SABC免疫组织化学法动态观察清营解表合剂干预治疗后肺组织细胞因子白介素-6(IL-6),白介素-10(IL-10)及肿瘤坏死因子-β(TNF-β)表达水平的变化。结果流感病毒感染小鼠后,感染模型组IL-6,IL-10及TNF-β表达各时间点均高于正常空白组,在第6天IL-6,TNF-β表达均达峰值,在第3,6天升高差异显著有统计学意义。药物干预后,清营解表合剂组IL-10表达逐渐升高,第6,9天较感染模型组升高差异显著。清营解表合剂组IL-6,TNF-β表达水平各时间点较感染模型组均下降,第6天下降差异有统计学意义。结论清营解表合剂通过调节流感病毒感染小鼠Th类细胞因子的表达水平而提高机体免疫功能,恢复机体的抗感染免疫平衡。 Objective To explore the immune mechanism of Qingying Jiebiao Heji in resisting influenza virus infection by dynamically observing its effect on the expressions of Th cytokine in lung tissue section of the mice infected by influenza virus. Methods Influenza virus(A/PR/8 /H1N1) infected mice was adopted as model and SABC immunohistochemistrial method was used to dynamically observe the changes of expression levels of IL-6,IL-10 and TNF-β in lung tissue section after giving Qingying Jiebiao Heji treatment. Results After the mice were infected by influenza virus,the expression levels of IL-6,IL-10 and TNF-β in model group was higher than those of control group ,of which IL-6,TNF-β reached the peak on the sixth day. After giving the treatment,the expression of IL-10 in Qingying Jiebiao Heji treated group was increased and the increase was remarkable on the sixth and ninth day,compared with those of model group. The expression levels of IL-6,TNF-β in Qingying Jiebiao Heji treated group was decreased in the different time and the decrease became remarkable on the sixth days,compared with those of model group. Conclusion Qingying Jiebiao Heji can increase the immune function and recover the immune balance of the mice infected by influenza virus by adjusting the expression level of Th cytokine.
