脾阳虚大鼠肽转运载体蛋白表达及转运功能的改变 被引量:7

摘要 目的观察脾阳虚模型大鼠小肠黏膜肽转运载体1(PepT1)的表达及其转运功能的变化。方法采用劳倦泄下合利血平复合因素造模法建立脾阳虚大鼠模型,RP-HPLC法测定cefalexin血药浓度以评价小肠黏膜PepT1的转运吸收功能,免疫组化方法观察脾阳虚大鼠肠上皮PepT1蛋白表达情况。结果十二指肠给药120min后,RP-HPLC法测定模型大鼠血清cefalexin浓度为(5.23±2.10)μg/ml,高于正常对照组的(2.52±0.47)μg/ml(P<0.01);模型大鼠小肠黏膜上PepT1蛋白表达较正常对照大鼠表达升高。结论脾阳虚证模型大鼠小肠黏膜肽转运载体PepT1表达及对cefalexin的转运能力增强。 Objective To observe the expression and transport ability of PepT1 protein in model rats of spleen-"yang" deficiency. Methods The model spleen-" yang" deficiency rats was established by the way of over exertion,purgation combined with reserpine injection. Transport ability of PepT1 for cefalexin was determined by RP-HPLC. Immunohistochemical technique was performed to detect the expression of PepT1 protein in mucous membrane of small intestine. Results After 120 minutes administration through duodenum injection,serum cefalexin concentration of model rats was ( 5. 23 ± 2. 10) μg/ml,higher than that of control group,( 2. 52 ± 0. 47) μg/ml. The PepT1 protein expression level in small intestine mucosa of model rats was higher than that of control group. Conclusion Compared to control group,the expression and transport ability of PepT1 in model rats of spleen-" yang" deficiency was increased.
