摘要
为了探讨蓝灰团网菌(Arcyria nigellaEmoto)的分子系统学的关系,对基物湿室培养获得的标本进行了rDNA的提取及ITS序列的扩增。首次通过玻璃棒研磨的方法对蓝灰团网菌DNA进行了提取,并得到了高质量的DNA。采用ITS引物进行PCR扩增得到了序列长度为587bp的rDNA片段,并在Genbank上进行了注册,登陆号为HM101140。同时,将该序列与大粉瘤菌(Lycogala flavofuscum)的相应序列(Genbank登陆号为AY743240)进行了比对,其序列同源性为70.6%。
rDNA was extracted from Arcyria nigella specimens obtained by moist chamber culture,and then the rDNA was amplified using ITS primers aiming to probe of the relationship between the molecular systematics of A.nigella.A new method,grinding with glass rod,for extracting DNA from myxomycetes was used to extract DNA for the first time,and high quality DNA was obtained.The rDNA sequence is 587bp in length,and it was registered in GenBank(Accession number:HM101140).Moreover,the sequence was aligned with Lycogala flavofuscum(Accession number:AY743240).The homology was 70.6%.
