三氧化二砷对人胆囊癌裸鼠移植瘤生长的抑制作用及其机制 被引量:3

Arsenic Trioxide Inhibits the Growth of Transplanted Tumor of Human Gallbladder Carcinoma and Its Mechanism in Nude Mice

摘要 目的:观察三氧化二砷(As2O3)对人胆囊癌裸鼠移植瘤生长的抑制作用并探讨其机制。方法:20只体质量相似的BALB/c裸鼠,皮下接种胆囊癌移植瘤随机分为0.9%氯化钠液(NS)组(n=10)和As2O3组(n=10)。尾静脉注射NS0.2mL(NS组)或As2O310mg.kg-1(As2O3组),每日1次,连续7d。21d时处死所有裸鼠,测量肿瘤的质量和体积,同时分别用免疫组织化学法和Western blot法检测细胞周期蛋白D1(Cyclin D1)的表达。结果:NS组瘤质量0.992±0.401g、体积1.50±0.57mm3,As2O3组瘤质量0.526±0.269g、体积0.83±0.39mm3,两组间均有显著差异(P<0.01);免疫组织化学和West-ern blot检测结果显示As2O3组肿瘤组织Cyclin D1的表达明显下降。结论:As2O3能明显抑制人胆囊癌裸鼠移植瘤的生长,可能通过下调Cyclin D1的表达来实现抑瘤生长。 Objective:To observe the inhibitory effect of arsenic trioxide(As2O3)on the growth of transplanted tumor of human gallbladder carcinoma and to probe into its mechanism in nude mice.Methods: Twenty BALB/c nude mice,with the resembling volume of subcutaneous transplanted tumor,were randomly assigned into eithor nature saline(NS) group or As2O3 groups.0.2ml NS or 10 mg·kg-1 As2O3 was injected into the tail vain of every mouse in NS group or As2O3 group,once a day,for seven days.All mice were put to death fourteen days after the perfusion was performed completely,then the weight and dimension were assayed as well as the expression of Cyclin D1 were determined by immunohistochemical staining and Western Blot.Results: The weight and volume of transplanted tumor was 0.992±0.401 g and 1.50±0.57 mm3,respectivey in NS group,which reduced to 0.526±0.269g and 0.83±0.39 mm3 respectivey in As2O3 group.The expression of Cyclin D1 also decreased significantly in As2O3 group.Conclusions: As2O3 can significantly inhibit the growth of transplanted tumor of human gallbladder carcinoma and its mechanism may be associated with down-regulation of expression of Cyclin D1.
