食管鳞癌中诱骗受体3与基质金属蛋白酶-2蛋白的表达及其临床意义

摘要 目的:探讨诱骗受体3(decoy receptor3,DcR3)和基质金属蛋白-2(matrix metalloproteinase-2,MMP-2)在食管鳞状细胞癌组织中的表达及其临床意义。方法:应用免疫组织化学方法检测76例食管鳞状细胞癌组织及30例癌旁正常组织中DcR3和MMP-2的表达情况。结果:食管鳞状细胞癌组织中DcR3蛋白、MMP-2蛋白阳性率分别为67.1%和51.3%,明显高于癌旁正常组织(分别为26.7%和16.7%)(P<0.01);两者在食管鳞状细胞癌组织表达具有正相关(r=0.271,P<0.05)。DcR3、MMP-2和DcR3/MMP-2在食管鳞状细胞癌组织中的阳性表达与癌组织分化程度、浸润深度、淋巴结转移均显著相关(P<0.05)。结论:高度表达的DcR3、MMP-2可能有助于食管鳞癌的发生、发展及早期侵袭转移,联合检测DcR3、MMP-2表达有可能作为反映食管鳞癌进展和预后的分子生物学指标。 Objective:To investigate the expression of decoy receptor 3(DcR3) and matrix metalloproteinase-2(MMP-2) in esophageal squamous cell carcinoma and adjacent normal mucosa,and the clinical significance.Methods:Immunohistoche-mistry technique was used to detect the expression of DcR3 and MMP-2 proteins in 76 cases of esophageal squamous cell carcinoma and 30 cases of adjacent normal mucosa.Results:The positive rates of DcR3 and MMP-2 protein expression were 67.1% and 51.3%,respectively,in esophageal squamous cell carcinoma,while only 26.7% and 16.7% in adjacent normal mucosa.The positive rates of DcR3 and MMP-2 protein expression in esophageal squamous cell carcinoma were significantly higher than those in adjacent normal mucosa(P0.01).The positively expression of DcR3,MMP-2 and DcR3/MMP-2 protein was significantly associated with differentiation of esophageal squamous cell carcinoma,depth of tumor in vasion and lymph node metastasis(P0.05).A positive correlaton was observed between DcR3 and MMP-2 protein in esophageal squamous cell carcinoma(r=0.271,P0.05).Conclusions:The overexpressed DcR3 and MMP-2 in esophageal squamous cell carcinoma may contribute to genesis,development and progression of the cancer.The join detecting DcR3 and MMP-2 may be a molecular-biological indicator in forecasting the progress and prognosis of esophageal squamous cell carcinoma.
