摘要
目的:观察六君子汤对幽门螺杆菌感染小鼠血清细胞因子IL-2、IL-8的影响,探讨六君子汤治疗幽门螺杆菌(Helicobacter Pylori,H.Py-lori)相关性胃炎的机制。方法:60只昆明种小鼠随机分为5组:正常对照组、模型组、六君子汤大、中、小剂量组,除正常对照组外,其余各组采用灌胃造模的方法建立H.Pylori相关性胃炎小鼠模型,六君子汤大、中、小剂量组分别给予相应六君子汤灌胃治疗,其余两组给予等体积的生理盐水灌胃。2周后,心脏采血处死小鼠,观察各组小鼠胃粘膜的形态学变化,参照悉尼分类系统进行评估。血液离心取上清,采用ELISA双抗体夹心法,检测血清中IL-2、IL-8的含量。结果:各组小鼠胃粘膜炎症评分比较:模型组与其余各组,尤其是正常组和六君子汤大剂量组比较差异显著,有统计学意义(P<0.05或P<0.01)。模型组小鼠血清IL-2含量下降,IL-8水平增高,与正常对照组比较差异有统计学意义(P<0.01)。六君子汤大剂量组IL-2水平较模型组明显增高,差异有统计学意义(P<0.05)。六君子汤大、中、小剂量组IL-8水平较模型组降低,其中六君子汤大、中剂量组与模型组比较差异有统计学意义(P<0.05或P<0.01)。结论:六君子汤通过升高H.Pylori相关性胃炎小鼠血清IL-2含量,降低血清IL-8水平,调整细胞因子间的网络平衡,改善机体免疫功能,减轻胃粘膜的炎症反应,从而发挥对胃粘膜的保护作用。
Objective: To observe the effect of Liujunzi Decoction on serum IL -2, IL -8 of rats infected by helicobacter pylori, and to explore the mechanism in the treatment of helicobacter pylori associated gastritis. Methods: 60 mice were randomly divided into 5 groups : normal control group, model group, Liujunzi Decoction groups of large, medium and small dose. Except for the normal control group, other groups were given gastric perfusion to establish model mice with H. Pylori associated gastritis; while the large, medium and low dose groups were given corresponding gastic perfusion of Liujunzi Decoction. the other two groups gastic perfusion of equal volume of normal saline. 2 weeks later, mice were killed to observe and evaluate the morphological changes of gastric mucosa with reference to the Sydney classification system. Supernatant as taken after blood centrifugation, and then detected serum IL - 2, IL - 8 levels by using double antibody sandwich ELISA. Results : The gastric inflammation score in different groups: model group and other groups, especially the normal group and the large dose group were significantly different, there was statistical significance ( P 〈 0.05 or P 〈 0. 01 ). Serum IL - 2 of model group decreased, while IL - 8 levels increased, compared with normal control group there was statistical significance ( P 〈 0.01 ). IL - 2 levels of Liujunzi Decoction group of large dose were significantly higher than those of the model group, and the difference was statistically significant ( P 〈 0.05 ). IL - 8 levels of large, medium and low dose group of Liujunzi Decoction were lower than those of the model group. Among them, compared with the large, medium dose group and the model group, the difference was statistically significant ( P 〈 0.05 or P 〈 0.01 ). Conclusion: Liujunzi Decoction may playa protective effect on the gastric mucosa through elevating serum IL -2 levels of mice with H. Pylori associated gastritis, lowering serum levels of IL - 8, adjusting the balance of cytokine network, improving immune function and reducing inflammation of gastric mucosa.
