右美托咪定对肺巨噬细胞炎性损伤的保护效应 被引量:19

摘要 目的观察右美托咪定(dexmedetomidine,Dex)对内毒素(LPS)联合γ-干扰素(IFN-γ)所致肺巨噬细胞活力损伤的影响。方法大鼠肺巨噬细胞NR8383接种于96孔培养板,分别采用正常培养液以及不同药物组合培养:100ng/mL LPS+50U/mL IFN-γ、100ng/mL LPS+50U/mL IFN-γ+10ng/mL Dex、100ng/mLLPS+50U/mL IFN-γ+1ng/mL Dex、100ng/mL LPS+50U/mL IFN-γ+10ng/mL Dex+10μmol/L Yohim-bine、10ng/mL Dex、1ng/mL Dex、10μmol/L Yohimbine(选择性α2肾上腺素能拮抗剂),24h后采用Western blot检测诱导性一氧化氮合酶(iNOS)生成量;采用四甲基偶氮唑盐比色(MTT)法检测细胞活力。结果 100ng/mLLPS联合50U/mL IFN-γ刺激引起NR8383细胞iNOS大量表达,细胞活力下降为正常培养细胞的47.8%。10ng/mL Dex可以明显抑制LPS联合IFN-γ刺激所引起的NR8383细胞iNOS表达量和细胞活力下降(63.4%vs47.8%,P<0.05);10μmol/L Yohimbine可部分阻断10ng/mL Dex的抑制效应;1ng/mL Dex对LPS联合IFN-γ刺激引起的NR8383细胞iNOS表达量和活力损伤没有明显影响。Dex、Yohimbine单独对NR8383细胞iNOS表达量和活力没有影响。结论 Dex(10ng/mL)能保护LPS联合IFN-γ所致肺巨噬细胞NR8383活力损伤,此效应是通过α2肾上腺素能受体介导的。
