摘要
目的研究P53、Caspase-3和增殖细胞核抗原(PCNA)蛋白在肝细胞癌中的表达和意义。方法采用免疫组织化学S-P法检测P53、Caspase-3和PCNA蛋白在69例肝细胞癌(HCC)及癌旁组织中的表达。结果 (1)P53和PCNA蛋白在肝细胞癌组织中的阳性表达率分别为56.52%、20.29%,在癌旁组织中的阳性表达率均为1.45%,肝细胞癌组织中P53和PCNA蛋白的阳性表达率显著高于癌旁组织,差异有统计学意义(P<0.05);(2)Caspase-3蛋白在肝细胞癌及癌旁组织中均有表达,差异有统计学意义(P<0.05);(3)P53蛋白的表达与肝细胞癌的分化程度呈负相关(rs=-0.239,P<0.05),肝细胞癌的分化程度越低,P53蛋白的阳性表达率及表达强度越高。结论 P53和PCNA蛋白的过度表达及Caspase-3蛋白的低表达可能在肝细胞癌的发生和发展中起重要作用。
Objective To investgate the expression and signifigence of p53,proliferating cell nuclear antigen (PCNA) and Caspase-3 proteins in hepatocellular carcinoma (HCC).Methods The expression of p53,PCNA and Caspase-3 protein were evaluated using immunohistochemistry in 69 HCC tissue and paracarcinoma tissue.Results (1) The positive rates of p53 and PCNA proteins in HCC were 56.52%,20.29%,the positive rates of p53 and PCNA in paracarcinoma tissue were both 1.45%,the positive rates of p53 and PCNA protein in HCC were higher than paracarcinoma tissue,with significant differences (P0.05);(2) Caspase-3 protein expressed in both HCC and paracarcinoma tissue,with significant differences (P0.05);(3) The expression of P53 protein was related to the differentiation degree of HCC (rs=-0.239,P0.05),the lower the differentiation degree of HCC was,the higher the expression rates and expression extent of P53 protein were.Conclussion Overexpression of p53,PCNA proteins and low expression of Caspase-3 protein may play a crucial role in the origin and development of HCC.
