副溶血性弧菌活的非可培养状态及其复苏菌株特性的分析 被引量:7

摘要 将副溶血性弧菌Vibrio parahaemolyticus BJ1.1997和Vibrio parahaemolyticus BJ1.1616分别于低温(4℃)寡营养(人工海水、陈海水)条件和低温(4℃)富营养条件(TSB-3%NaCl)下进行诱导实验。结果显示,两菌株皆可进入活的非可培养状态(Viable but Non-Culturablestate,VBNC),且进入VBNC状态的菌株对小白鼠无致病性。运用升温复苏等多种方法对已经进入VBNC状态的副溶血性弧菌进行复苏,并对复苏菌株的生长曲线、生理生化指标和毒力基因进行测定。结果表明,部分菌株复苏成功,复苏后菌株的生理生化指标和毒力基因与标准菌株基本相同,但V.pA7F(Vibrio parahaemolyticus BJ1.1997复苏菌株)的生长活性有所下降,并且其trh毒力基因和脲酶反应均为阴性。以上结论皆为研究副溶血性弧菌VBNC和食品安全控制提供理论依据。 This article is about Vibrio parahaemolyticus BJ1.1997 and Vibrio parahaemolyticus BJ1.1616 which were respectively induced in oligotrophic(Artificial seawater,Seawater) conditions and eutrophic conditions(TSB-3% NaCl) at low temperatures(4 ℃).The results showed that both of them could reach the viable but nonculturable(VBNC) state.And this VBNC state of strains is non-pathogenic on mice.Different methods,such as temperature upshift etc,were used for resuscitating from this state.Growth curves,physiological and biochemical indexes and virulence genes of resuscitated strains were detected.The results showed that some strains could resuscitate successfully.The physiological and biochemical indexes and virulence genes of resuscitated strains were the same as standard strains.But the growth activity of resuscitated V.p A7F(resuscitated in Vibrio parahaemolyticus BJ1.1997)strains had a slight decrease.The urease and virulence gene for trh had a negative reaction.This study provided the theoretical basis for the research of Vibrio parahaemolyticus in VBNC state and food safety control.
