摘要
[目的]建立了同时测定虎杖(Polygonum cuspidatum Sieb.et Zucc.)提取物中白藜芦醇、虎杖苷、大黄素和大黄素甲醚含量的HPLC法。[方法]采用DIKMAC18色谱柱(200.0mm×4.6mm,5μm),以乙腈-0.1%磷酸溶液为流动相,流速为1.0ml/min,波长为303nm,柱温为30℃,进样量为10μl。[结果]白藜芦醇、虎杖苷、大黄素和大黄素甲醚对应的回归方程分别为y=7116169x+15631(r=0.9999)、y=395313x+50(r=0.9999)、y=2752869x-2664(r=0.9999)、y=463886x-1413(r=0.9999),且白藜芦醇在0.026~0.520μg范围内、虎杖苷在0.045~0.765μg范围内、大黄素在0.026~0.520μg范围内、大黄素甲醚在0.026~0.442μg范围内线性关系良好。[结论]该研究建立的方法可行,重复性好,可用于虎杖药材的质量控制。
Objective] The research aimed to establish a method for the content determination of resveratrol,polydatin,emodin and physcion in Polygonum cuspidatum Sieb.et Zucc. simultaneously by HPLC. [Method] HPLC method was established with DIKMA C18 column(200.0 mm×4.6 mm,5 μm)and a mobile phase of acetonitrile-0.1% phosphoric acid. The flow rate was 1.0 ml/min,the detection wavelength was 303 nm and the column temperature was 30 ℃. [Result] The regression equations of resveratrol,polydatin,emodin and physcion were y=7 116 169 x + 15 631(r=0.999 88),y=395 313 x + 50,y =2 752 869 x -2 664 and y = 463 866 x -1 413 respectively. Resveratrol,polydatin,emodin and physcion showed a good linear relationship in the range of 0.026-0.520,0.045-0.765,0.026-0.520 and 0.026-0.442 μg respectively. [Conclusion] The established method is feasible and the repeatability is good. The method can be used for quality control of P. cuspidatum medicine materials.
