快速荧光灶抑制试验对我国部分肾综合征出血热毒株及病人血清的分型研究

Studies on Serotypic Classification of Viruses of Hemorrhagic Fever with Renal Syndrome and Patient’s Sera by Rapid Fluorescent Focus Inhibition Test

目的：探讨肾综合征出血热（ＨＦＲＳ）毒株及病人血清分型方法。方法：快速荧光灶抑制试验（ＲＦＦＩＴ）。结果：６株来自黑线姬鼠和病人的毒株，其免疫血清对Ⅰ型标准株７６－１１８的中和效价较Ⅱ型标准株（ＵＲ）高４～２５６倍；而另６株来自褐家鼠和大白鼠的毒株，包括Ｇｏｕ３和Ｋ２４的免疫血清，对Ⅱ型标准株ＵＲ的中和效价高于Ⅰ型７６－１１８株８～１２８倍；被检的所有毒株和病人血清都能准确地分型。结论：ＲＦＦＩＴ具有快速、简便、特异和准确灵敏的特点，不仅可检测中和抗体，且适用于ＨＦＲＳ病毒血清学分型。

Serotypic classification of 12 HFRS viruses isolated from different sources and patient sera from different areas was studied using rapid fluorescent focus inhibition test (RFFIT).Sera of rabbits inoculated with different HFRS viruses were tested against reference type-ⅠHantaan virus and type-Ⅱ Seoul virus .The neutralizing titers of antisera of Apodemus isolates and patient blood against type-Ⅰ Hantaan virus were 4～256 fold higher than those to type-Ⅱ seoul virus,while the antisera of Rattus isolates-had titers to type-Ⅱ viruses 4～128 fold higher than those to type-Ⅰ viruses.So different isolates could be classified into serotype-Ⅰ and serotype-Ⅱ.In this study,we also found Gou 3 and K 24 ,which could be classified into serotype Ⅱ according to differences (≥4-fold) of the neutralizing titers between the sera reacted against reference type-Ⅰ and type-Ⅱ viruses, which is consistent with data previously published.Otherwise, serotypic classification of patient sera from different areas was investigated. The results showed that the RFFIT is an easy, fast、 specific and exact method and may be used in Serotypic classification of HFRSV.
