摘要 目的探讨TLR9在胃癌细胞MGC803上的表达及其激动剂对MGC803细胞增殖和凋亡的影响。方法用RT-PCR方法检测MGC803细胞上TLR9的表达;用TLR9的激动剂CpG寡脱氧核苷酸(CpG oligodexy-nucleotides,CpG-ODN)10μg.mL-1分别作用MGC803细胞24、48 h和72 h后,用MTT法检测细胞生长抑制率;流式细胞术观察CpG ODN作用MGC803细胞24 h后的细胞凋亡现象。结果 MGC803细胞上有TLR9的表达;TLR 9激动剂CpG ODN作用于MGC803细胞不同时间点,均能明显抑制MGC803细胞生长(P<0.01),以早期凋亡为主。结论 TLR9激动剂CpG ODN对胃癌细胞的生长具有抑制作用。 Objective To investigate the expression of TLR9 in the human gastric cancer cells and the effects of TLR9 agonist CpG-ODN on proliferation,apoptosis of MGC803 ce lls.Methods To measured the expression of TLR9 in MGC803 cells by RT-PCR;Observed the inhibitive effect of TLR9 agonist（CpG-ODN）10 μg·mL-1 in MGC803 cell at 24 h、48 h、72 h by MTT assay;apoptosis were detected by f low cytometry（FCM）.Results MGC803 cells expressed TLR9;TLR9 agonist CpG-ODN could inhibit the growth of MGC803 cells（P0.01） at different time point,and the cell treated with CpG-ODN are in the early stage of apoptosis.Conclusion TLR9 agonist CpG-ODN may be able to inhibit the growth of MGC803 cells.
