摘要
从基因工程水平构建了小鼠金属硫蛋白与抗人活化血小板单抗SZ51单链抗体的重组基因产物,并在大肠杆菌菌株BL21(DE3)pLysS中成功地进行了表达.该重组蛋白以包涵体形式存在于菌体蛋白中,分子量为38kD,ELISA实验证明该重组蛋白既具有血栓部位活化血小板的单抗活性。
With genetic engineering methods,a kind of mouse metallothionein and single chain antibody of human activated platelet antibody SZ51 recombinant product was constructed and successfully expressed in the E.Coli BL21(DE3)pLysS strain.The reult shows,the recombinant protein existed in the form of inclusion body in the total strain proteins,and the molecular weight of the recombinant protein is 38 kD,after the enzymatic linked immuntity absorbence test,it shows that the recombinant protein not only have the ability of the activity of anti human activated platelet the same as the previous antibody SZ51 but also have the activity of anti mouse metallothionein the same as the native metallothionein.This study will provide with new,effective directing and imaging agent in the field of thrombus determination.
