摘要
目的探讨细胞周期依赖蛋白激酶2(CDK2)及细胞周期蛋白依赖激酶抑制因子(P27)在高氧致慢性肺疾病(CLD)新生鼠肺纤维化中的动态表达规律及作用。方法采用高浓度氧诱导新生鼠CLD模型,应用免疫组织化学法、实时定量PCR法及Western blot技术,检测肺组织CDK2及P27蛋白和基因的表达,并同时测定肺组织纤维化评分。结果与对照组相比,实验组14、21dCDK2蛋白和基因的表达明显升高(P<0.05),P27的表达明显降低(P<0.05)。CDK2的动态表达变化与纤维化程度呈正相关(r=0.702),P27与纤维化程度呈负相关(r=-0.765)(P<0.05)。结论 CDK2及P27的异常表达与高氧致CLD肺纤维化密切相关。
Objective To investigate the expression and role of cyclin-dependent kinase-2(CDK2)and cyclin-dependent kinase inhibitors (P27)in pulmonary fibrosis in neonatal rats with hyperoxia-induced chronic lung disease (CLD).Methods CLD model was induced by hyperoxia exposure.The expression of CDK2 and P27 protein and mRNA were detected by immunohistochemiscal method,Real-time PCR and Western blot.Results Compared with the control group,the expression level of CDK2 protein and mRNA were increased significantly (P 〈 0.05)on day 14 and 21,while the expression of P27 were decreased significantly (P 〈 0.05).The dynamic expression of CDK2 was positively correlated with the degree of lung fibrosis,and the dynamic expression of P27 was negatively correlated with the degree of lung fibrosis.Conclusion The abnormal expression of CDK2 and P27 were related to pulmonary fibrosis in neonatal rats.
