摘要
目的:运用蛋白质组学的理论和方法来研究中小强度有氧运动对大鼠右心室肌蛋白质组的影响。方法:将20只SD雄性大鼠随机分为对照组和运动组(各1组,每组n=10)。采用中小强度运动负荷跑台训练建立运动实验模型,提取右心室肌组织全蛋白样品,采用双向凝胶电泳进行蛋白样品分离。运用ImageMaster 2D Platinum图像分析软件对2-DE胶进行分析,通过图像分析,选取运动后差异表达量≥5倍的蛋白点,用串联飞行时间质谱蛋白质仪(ULGRAFL-FLEX-TOF/TOF)进行质谱鉴定。结果:经过8周运动后,在2-DE图谱上运动组和对照组检测到的蛋白质点分别为(547±18)个和(530±14)个,匹配率分别为(71.34±3.25)%和(70.28±2.38)%。运动后共有58个蛋白质点发生了变化:表达量上调≥2倍的点有33个,下调≥2倍的点有25个,其中上调≥5倍的点有8个,下调≥5倍的点有8个。选择差异表达量≥5倍的蛋白质点为目标蛋白质进行质谱分析,共鉴定出了5个差异表达蛋白质点。结论:在长期中小强度有氧运动后,大鼠右室肌蛋白质组发生了明显的差异性表达,这些差异性表达的蛋白质点为进一步观察在运动应激状态下对心脏结构与功能影响的新的目标蛋白质提供了实验依据。
objective:Proteomic methodology was used to study the influence of Moderate and small intensity aerobic exercise on the right ventricle proteins in rats.Methods:20 rats were divided into rest and exercise groups in random.The exercise group was set up to do treadmill training with medium and small intensity exercise and then killed to extract the total protein of right ventricular muscle for two dimensional gel electrohoresis searation.The spots in different expression volume more than 5 times are selected as alternative spots for mass sectra(MS) identification.The gels maps were analyzed with the ImageMaster 2D Platinum image analysis software to choose variable proteins and the chosen proteins were identified by ULGRAFL-FLEX-TOF/TOF.Results:After 8 weeks' training,in electrophoresis map,the average number of detected protein spot in rest group and exercise group are respectively 530±14 and 547±18,and the match ratio are respectively(70.28±2.38)% and(71.34±3.25)%.There are 58 changed after exercise:33 increased expression more than 2 times;25 decreased expression more than 2 times.Protein spots of differential expression more than 5 times were selected as object proteinum in MS identification and 5 protein spots of which were assess.Conclusion:right ventricle protein were obviously variously expression after long term moderate and small intensity aerobic exercise,it provides sufficient experimental bases in its further study.
