摘要
目的建立用高效液相色谱法(HPLC)测定小花黄堇中主要有效成分延胡索乙素含量的方法。方法色谱条件为色谱柱Gemini C18柱(250 mm×4.6 mm,5μm);流动相为甲醇-0.1%磷酸溶液(75∶25);流速1.0 mL/min;检测波长280 nm;柱温25℃。结果小花黄堇中延胡索乙素的平均含量为0.160%,延胡索乙素在0.193~2.128μg范围内线性关系良好。平均回收率为98.55%,RSD为1.67%(n=6)。结论该方法操作简便,重复性较好,测定结果较为可靠,可用于测定小花黄堇中延胡索乙素的含量。
Objective To establish a method for quantitative determination of tetrahydropalmatine in Xiaohuahuangjin(Corydalis racemosa) by HPLC.Methods The HPLC procedure was performed on the chromatographic column of Gemini C18(250 mm×4.6 mm,5 μm) at 25 ℃,the mobile phase was methanol-0.1% phosphoric acid water solution(75∶25).The flow velocity was 1.0 mL/min and the detection wavelength was 280 nm.Results The average content of tetrahydropalmatine in Xiaohuahuangjin was 0.160%.Tetrahydropalmatine showed a good linear relationship at a range from 0.193 μg to 2.128 μg.The average recovery was 98.55%(RSD=1.67%,n=6).Conclusion The method was simple and reliable with a good repeatability,which can be used for the quantitative determination of tetrahydropalmatine in Xiaohuahuangjin.
