摘要
目的:研究猴头菌液体培养提高菌丝生物量的最佳配方。方法:通过对猴头菌液体发酵的各常用碳源与氮源单因素试验和正交试验L_9(3(?)4),研究了葡萄糖、玉米粉、淀粉、蔗糖、酵母粉、蚕蛹粉、蛋白胨、黄豆粉对提高菌丝生物量的影响。结果:葡萄糖、玉米粉复合碳源为最适碳源,蛋白胨、黄豆粉为最适氮源,从而得到了猴头菌液体发酵培养的最佳培养基为:葡萄糖30g/L、玉米粉30g/L、蛋白胨4g/L、黄豆粉15g/L、KH_2PO_41.5g/L、MgSO_40.75g/L;培养条件为:温度25℃,时间为(140~144)h,在最佳培养工艺条件下,猴头菌丝体的收率为28.1g/L,比基础工艺的菌丝收率提高了一倍多,菌丝体中的多糖、多肽含量分别为基础工艺的1.34倍与1.12倍。结论:通过正交试验得到的培养基配方菌丝生物量收率高,有效成分多糖、多肽含量高,工艺稳定,可作为猴头菌液体发酵的生产工艺。
Objective:To study the liquid culture of Hericium erinaceus mycelium biomass to improve the best formula.Methods: Fermentation Hcricium commonly used carbon and nitrogen in various single-factur experiments and orthogonal test L9 (3^4),of glucose,maize flour, starch,sugar,yeast powder, silkworm chrysalis powder,peptone,soybean flour to improve the mycelium biomass.Results:glucose,corn flour as carbon sources for the optimal carbon source,peptone,soybean powder The Optimal nitrogen source,which has been submerged cultivation of Hericium the best medium: glucose 30g/L,com flour 30g/L,peptone 4g/L,soy flour 15g/L,KH2PO4 l.5g/L,MgSO4 0.75g/L;cultural conditions were: temperature 25℃,time for the 140 -144 h,the optimum culture conditionsithe Hericium mycelium yield was 28.1g/L,based on technology than the yield of mycelia has doubled and more mycelium polysaccharide,polypeptide content-based technique was 1,34 times and 1.12 times: Conclusion:get the medium orthogonal formula mycelium biomass yields and effective component of polysaccharidelpolypeptide content was high,process stability, as Hericium liquid femlentation production process.
