通利大肠对慢性阻塞性肺疾病大鼠肺组织γ-GCS及Nrf2 mRNA表达的影响 被引量:4

Influences of relaxing large intestine therapy on γ-GCS, Nrf2 mRNA expression in rats with COPD

摘要 目的:观察"通利大肠"对慢性阻塞性肺疾病(COPD)模型大鼠肺组织γ-谷氨酰半胱氨酸合成酶(γ-GCS)、核因子-2-相关因子-2(Nrf2) mRNA表达的影响,探讨COPD"从肠论治"效应产生的肺肠联络机制。方法:采用气管注脂多糖加熏香烟联合造模方法建立COPD大鼠模型,随机分为正常组、模型组、治肺组、治肠组及肺肠同治组。正常组、模型组灌胃生理盐水,各给药组灌胃相应中药,连续14d。RT-PCR法检测肺组织γ-GCS、Nrf2 mRNA表达水平。结果:与正常组比较,模型组大鼠肺组织γ-GCS、Nrf2 mRNA表达明显增强(P<0.01)。与模型组比较,各用药组γ-GCS、Nrf2 mRNA表达减少(P<0.01)。与治肺组比较,肺肠同治组γ-GCS、Nrf2 mRNA表达减少(P<0.01)。结论:通利大肠,或在治肺的基础上增加通利大肠,均能减少COPD大鼠肺组织γ-GCS、Nrf2mRNA的表达,这可能是COPD"从肠论治"效应产生的作用环节之一。 Objective: To observe the influences of relaxing large intestine therapy on γ-GCS, Nrf2 mRNA expression in rats with COPD, and to investigate the mechanism of relaxing large intestine therapy. Methods: The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of LPS. All rats were randomly divided into normal control group, model group, treating lung group, treating intestine group, treating lung and intestine group. The normal control group, model group were given intragastrically the normal saline solution, and other groups were given corresponding herbal drugs intragastrically for 14 days. Using real-time PCR to detect expression of γ-GCS, Nrf2 mRNA. Results: The levels of γ-GCS, Nrf2 mRNA expression were markedly higher in the model group, compared with those in the normal control group (P0.01). γ-GCS, Nrf2 mRNA expression was reduced in the treating intestine group than those in model group (P0.05). Moreover, γ-GCS, Nrf2 mRNA expression was also reduced in the treating lung and intestine group than those in the treating lung group (P0.01). Conclusion: relaxing large intestine therapy can reduce the expression of γ-GCS, Nrf2 mRNA.
