气虚证大鼠肾上腺上调基因的筛选

Up-regulated expression genes in adrenal gland of rats with Qi deficiency syndrome

摘要 目的:筛选气虚证大鼠肾上腺上调的基因。方法:采用16周龄Wistar大鼠、自发性高血压大鼠(SHR)、自发性糖尿病(GK)大鼠,采用标准化大鼠诊法,及GeneChipRatGene1.0STArray等技术,检测Wistar正常与气虚大鼠、SHR气虚和气盛大鼠、GK气虚和气盛大鼠肾上腺基因表达谱,筛选3个气虚证肾上腺一致上调的基因。结果:①获得3个气虚证同时上调的高表达基因15个,主要包括参与代谢、运输类的Cyp11b2、Fdxr、Gpd1、Abcc5、Kcnk9、Slc6a5、Slc6a6,参与信号转导类的Cd97、Calm3、Mapk14、Uba1、Sfrs2。②获得3个气虚证同时上调中等表达基因25个,包括参与代谢、运输类的Akt1、Usp19、Pex14、Slc27a1、Slc4a3、Tcirg1,参与转录调控及信号转导类的Pkn1、Gatad2a、Crebl1、Notch2、Adrbk1、Grk6、Bcar1、Myo9b等。③部分生物学功能不明确的基因,包括Trim25、Gpsn2、Itm2c、Pvrl2、Scyl1、Fgfr1、Cd99l2、Dclk2等。结论:气虚证Wistar、SHR与GK大鼠肾上腺组织上调表达的基因主要涉及代谢、运输、调控转录、信号转导等生物学过程的紊乱,其中与类固醇激素合成密切相关的Cyp11b2、Fdxr基因可能参与疾病及其气虚证的发生。 Objective: To screen and analyse up-regulated genes in adrenal gland of rats with qi deficiency syndrome. Methods: 16-week-old Wistar rats, spontaneously hypertensive rats (SHR) and Goto-Kakizaki (GK) Wistar rats were experimented, by the quantitative four diagnosis and syndrome differentiation methods and GeneChip Rat exon 1.0 ST Array, we observed adrenal gland gene expression about normal Wistar rats, qi deficiency Wistar rats, SHRs with qi deficiency and qi vigorous, GK rats with qi deficiency and qi vigorous. Up-regulated genes in three qi deficiency syndrome rats were analyzed. Results: Fifteen up-regulated and high expression genes were gained in three qi deficiency syndrome rats. Those genes were involved in metabolism and transport biological process, such as Cyp11b2, Fdxr, Gpd1, Abcc5, Kcnk9, Slc6a5 and Slc6a6, and involved in signal transduction biological process, such as Cd97, Calm3, Mapk14, Uba1, Sfrs2. Twenty five up-regulated and medium expression genes were gained in three qi deficiency syndrome rats. It included metabolism and transport biological process genes such as Akt1, Usp19, Pex14, Slc27a1, Slc4a3 and Tcirg1, and including transcription regulation and signal transduction genes such as Pkn1, Gatad2a, Crebl1, Notch2, Adrbk1, Grk6, Bcar1, Myo9b and so on. Some genes biological function was not clear, for example, Trim25, Gpsn2, Itm2c, Pvrl2, Scyl1, Fgfr1, Cd99l2, Dclk2 and so on. Conclusion: Up- regulated genes about qi deficiency Wistar, SHR and GK rat adrenal gland mainly related to the disorder of biological processes such as metabolism, transport, regulation of transcription, signal transduction. Moreover, Cyp11b2 and Fdxr may be related to hypertension and diabetes and qi deficiency syndrome.
