Regulation of Pregnane X Receptor (PXR) by Herbal Medicines

孕烷X受体(Pregnane X Receptor,PXR)是转录调控因子,参与大量的外源性和内源性化学物质的生物转化,这些物质都能激活PXR如胆汁酸、胆红素、某些维生素等。PXR还参与各种生理/病理过程,如脂代谢、糖代谢和炎症。PXR的配体结构多种多样,越来越多的患者在接受西药治疗的同时服用中药作为替代补充治疗的手段。中药的合用增加了临床上中西药发生相互作用和不良反应的风险,导致病患对化疗药物疗效不佳或产生毒性反应。本文主要关注中药对PXR的调节作用。

Pregnane X receptor （PXR） is transcription factors which control the expression of a broad array of genes. PXR regulated not only in transcellular transport and biotransformation of many drugs, other xenochemicals, and endogenous substances, such as bile acid, bilirubin, and certain vitamins, but also in various physiological/pathophysiological processes such as lipid metabolism, glucose homeostasis, and inflammation. Ligands of PXR are chemicals of diverse structures, including naturally occurring compounds present in herbal medicines. An increasing number of patients are using herbal medicines in combination with their conventional chemotherapeutic treatment. Herbal medicines increase the risk of clinically relevant herb-drug interactions, therefore leading to substantially more unex- pected toxicities of chemotherapeutic drugs and possible undertreatment seen in patients.The aim of this article is to provide an overview of the role of herbal medicines as modulators of PXR.
