脑忆源胶囊中黄酮类物质含量测定

Determination of Total Flavonoids in Naoyiyuan Capsules

【目的】建立脑忆源胶囊中总黄酮和淫羊藿苷含量的测定方法。【方法】采用《保健食品检验与评价技术规范》(2003年版)中规定的方法,以芦丁为对照品,采用紫外—可见分光光度法测定总黄酮含量,高效液相色谱(HPLC)法测定胶囊中淫羊藿苷含量,色谱柱:luna Cl8(2)100R(150 mm×4.6 mm,5μm),流动相:乙腈—水(体积比为30∶70),流速:1.0 mL/min,检测波长:270 nm。【结果】芦丁在0～24.75μg/mL范围内呈良好的线性关系,r=0.999 0,平均加样回收率为102.29%,sR为1.34%(N=5);淫羊藿苷含量在6.9×10-2～4.83×10-1μg范围内与峰面积积分值呈良好线性关系,r=0.999 9,平均回收率为95.39%,sR为2.5%(N=5)。【结论】本研究所建立的方法能够准确测定脑忆源胶囊中黄酮类成分的含量。

Objective To establish a method for the determination of total flavonoids and icariine in Naoyiyuan Capsules. Methods With rutin as reference, total flavonoids was determined by uhraviolet-speetrophotometer with the method which was recorded in Health Food Test and Evaluation Technological Specification （2003 edition）. The content of icariine in Naoyiyuan Capsules was detected by HPLC. The chromatographic separation was performed on Luna C18 （2） 100R column （ 150mm x 4.6mm, 5μm） with mobile phase consisted of acetonitrile-water （30： 70） at a flow rate of 1.0 mL/min. The detection wavelength was set at 270 nm. Results Rutin showed a good linear relationship at the range of 0-24.75 μg,/mL, r = 0. 9990, and the average recovery was 102.29% with RSD being 1.34%. Icariine had good linearity in the concentration range of 6.9×10^-2～4.83×10^-1 μg, r =0. 9999. The average recovery was 95.39% with RSD being 2.5% （N =5）. Conclusion The method is simple and accurate, and it can be used for the determination of total flavonoids in Naoyiyuan Capsules.
