22个毛木耳菌株的ISSR分析 被引量:12

摘要 采用ISSR分子标记技术对22个毛木耳菌株进行分析,应用NTSYSpc2.1生物软件进行遗传聚类,构建系统树。试验筛选出12个扩增谱带清晰、多态性好的ISSR引物,共扩增出80条清晰易辨的多态性谱带。聚类分析结果表明,利用ISSR技术可将22个毛木耳菌株材料区分开,遗传相似系数变异范围为0.100-0.846,遗传差异较大,在相似系数为0.421时,22个菌株聚为聚为4个类群,其中在四川广泛栽培的主栽品种黄耳10号、琥珀、781之间存在较大遗传差异。 22 Auricularia polytricha strains were studied using ISSR( Inter-Simple Sequence Repeat) method and the dendrogram was generated by UPGMA (unweighted pairgroup method with arithmetic average)method based on the similarity coefficient of ISSR patters. 12 ISSR primers were screened from 60 tested ISSR primers and 80 distinct and polymorphic DNA bands of tested strains were amplified. Clustering results revealed that 22 accessions could be distinguished using ISSR method, and similarity coefficients ranged from 0. 100 to 0. 846. All accessions were clustered into 4 groups on the similarity of 0. 421, and 3 major varieties in Sichuan province, including Huang erl0#, Hupo and 781 ,had a large genetic distance.
