摘要
通过培养基配方筛选和接种方法研究,得出金耳有效菌种制备的最佳母种和原种(栽培种)配方,及最佳的制种接种方法——组织块接种法,并总结出金耳有效菌种制备的技术规范。最佳母种配方:马铃薯(去皮)200 g,磷酸二氢钾3 g,蔗糖30 g,硫酸镁1.5 g,琼脂20 g,水1000 mL,pH自然;制备金耳有效优质原种(栽培种)最佳配方:中药渣90%,玉米粉6%,蔗糖2%,磷酸二氢钾1%,石膏1%。采用不同的接种方法,筛选出最好的接种方法为组织块接种法。
The composition of media for stock culture and spawn and the inoculating technique of Tremella aurantialba were studied. The optimal medium for stock culture was PSA with 3g KH2PO4 ,1.5g MgSO4,30g sucrose and 20 g agar in 1000mL 20 % popato juice. The optimal medium for spawn consisted of 90 % Chinese medicine residue,6 % corn powder,2 % sucrose,2 % KH2PO4, 1% Gypsum. The optimal inoculum was tissue piece. The standard programme for good spawn preparation was summarized.
