东方百合“索蚌”离体培养快繁体系建立 被引量:8

摘要 本研究以东方百合"索蚌"为试材,以MS培养基为基础培养基,附加不同种类和浓度的植物生长调节物质(6-BA,NAA,IBA)诱导丛生芽及再生植株。结果表明:"索蚌"的最适灭菌时间为8-10 min,最佳取材部位是鳞片的基部。东方百合"索蚌"的最佳诱导分化培养基为MS十6-BA2.0+NAA0.1,最佳继代培养基为MS+6-BA 2.0+NAA 0.2;最佳生根培养基为1/2MS+NAA0.1+IBA0.01,生根率97.8%,平均每苗根数可达11条;试管苗移栽的最佳基质为珍珠岩∶草炭土∶河沙=1∶1∶1,成活率达92%。 The influence of different concentration hormone (6-BA, NAA, IBA) for the tissue culture of oriental lily ' Sorbonne' , based on MS medium were studied in order to explore the suitable level of hormone and to increase the efficiency of the tissue culture of oriental lily ' Sorbonne' , and the different components of the four medium were used to find the best medium and best growth conditions, and transplanting of oriental lily' Sorbonne' was studied to establish the high frequency propagation system oriental lily' Sorbonne'. The best sterilization method was that shoot tip was soaked in the 75 % of alcohol for 30 s, and then put in 0.1% mercuric chloride for 8 - 10 min; the best drawn parts was the base of bulbs. Enrich of MS medium with MS +6-BA 2.0 + NAA 0.1 was the best to promote the induction of adventitious shoots and their growth, the optimal subculture medium was MS + 6-BA 2.0 + NAA 0.2 ,the optimum rooting medium was I/2MS supplemented with NAA0. 1 + IBA0. 01, rooting rate was 97.8 %, and Oriental lily's average each seedling radical .could reach 11. The transplanting survival rate was high on the transplanting medium of pearlite: vermiculite: peat: sand = 1: 1: 1: 1, the survival rate was up to 92 %.
