摘要
目的 探讨细胞色素P450表达与妊娠高血压的关系。方法比较下调细胞色素P450表达对妊娠晚期血压以及对胎鼠发育的影响,运用有创血压测定,胎鼠和胎盘发育指标以及westernblot,评价细胞色素P450表达与胎鼠发育的关系。结果与对照组相比,抑制细胞色素P450之后,实验组血压明显升高(118.6±8)mmHgvs(95.1±7)mmHg,P〈0.05),实验组的胎鼠重量、身长和胎盘的重量都较对照组显著降低(1.8±0.39)gvs(2.76±0.61)g,(1.78±0.32)VS(3.04±0.58)cm,(0.38±0.12)VS(0.45±0.15)g,P〈0.05),同时实验组胎盘中CYP2C23表达显著下降。结论抑制胎盘CYP2C23表达可能是导致胎鼠发育受限制的原因,下调细胞色素P450表达是妊娠高血压的重要发病机制。
Objective To study the relation of cytochrome P450 expression and pregnancy induced hypertension. Methods Compared the changes after inhibition of cytochrome P450 expression, to evaluate the function of cytochrome P450 in pregnancy induced hypertension with blood pressure measure, fetus development and western blot. Results Comparing with control group, the blood pressure increase ( 118.6 ± 8 ) vs ( 95.1 ± 7 ) mm Hg, P 〈 0. 05 ) , the weight and length of fetus, and the weight of placenta decrease ( 1.8±0. 39 ) vs ( 2. 76±0.61)g,(1.78 ±0.32)vs(3.04±0.58)cm,(0.38±0.12)vs(0.45±0.15)g,P〈0.05),western blot showed the expression of CYP2C23 was inhibited by MSPPOH in plancta. Conclusion These results demonstrated the inhibition of CYP2C23 expression was the cause of fetus retardation. Downregulation of cytochrome P450 expression might be the mechanism of pregnancy induced hypertension.
