摘要
为研究Fascin-1表达对喉鳞癌细胞骨架结构和运动能力的影响,采用RNA干扰技术抑制喉鳞癌细胞Hep-2中Fascin-1的表达,在激光共聚焦显微镜下观察Fascin-1表达下调对Hep-2细胞骨架结构的影响,并通过Millicell小室法检查Hep-2细胞的体外迁移能力。研究结果发现抑制Fascin-1蛋白在Hep-2细胞中的表达后,Hep-2细胞骨架结构的完整性被破坏,同时抑制了Hep-2细胞的迁移能力,Fascin-1蛋白表达下调组Hep-2细胞迁移抑制率为44.6%±6.3%,与对照组相比差异有统计学意义(P<0.05)。由此可推论,Fascin-1的表达与Hep-2细胞骨架结构的完整性有关,可能参与了Hep-2细胞体外运动的过程。
This study sought to detect the effect of Fascin1 expression on the cytoskeleton and immigration of laryngeal squamous carcinoma cell.In the experiment,Fascin-1 expression in Hep-2 cells was inhibited by small interfering RNA.The cytoskeleton of Hep-2 cells was observed with the use of laser scanning confocal fluorescence microscope.Millicell insert was applied to detect the immigration of Hep2 cells in vitro.The results showed that the integrity of cytoskeleton in Hep2 cells was broken with the down-regulation of Fascin-1 expression and the immigration ability was decreased significantly(P〈0.05).The inhibiting ratio of cell immigration was 44.6±6.3%.In conclusion,inhibition of Fascin-1 expression in Hep-2 cells could break the integrity of cytoskeleton and decrease the ability of cellular immigration.
