摘要
根据α-氨基酸和茚三酮在一定条件下发生显色反应的原理,研究了茚三酮比色法测定赖氨酸含量的方法,探讨了溶液pH、显色剂用量、加热时间和冷却时间对茚三酮比色法定量检测赖氨酸的影响,并测定了赖氨酸标准曲线。结果表明:溶液pH为6.0,显色剂用量为1.0mL,加热15min,冷却10min,波长570nm处测定吸光度为茚三酮比色法测定赖氨酸含量的最佳检测条件。
According to the colorimetry principle of α-amino acid and ninhydrin,the method of quantitative determination of lysine by ninhydrin colorimetry was researched,and the pH value of the solution,dosage of the ninhydrin,heated time and cooled time were investigated in order to find the optimum conditions of determination of lysine by ninhydrin colorimetry.The optimun conditions were as follows:the pH value was 6.0,and the ninhydrin was added 1.0mL,heated 15min and cooled 10min,finally determined at 570nm.
