摘要 目的观察咪唑安定对斑马鱼胚胎血管发育的影响。方法根据培养液成分将特异性标记血管内皮细胞的转基因斑马鱼胚胎随机分为四组,每组120个。A组咪唑安定浓度1mg/L、B组10mg/L、C组20mg/L、D组不加咪唑安定作为对照。咪唑安定处理时间为胚胎受精后4.3~30h,随后用共聚焦显微镜观察斑马鱼胚胎血管发育情况。结果与D组比较,A组斑马鱼胚胎无明显血管发育改变;C组胚胎发育停滞在原肠胚时期;B组胚胎体节间血管和尾部血管发育均受到明显的抑制(P<0.01)。结论 10mg/L的咪唑安定干预可抑制斑马鱼胚胎血管发育。 Objective To investigate the effects of midazolam on zebrafish embryonic vascular development. Methods The transgenic embryos treated with different concentrations of nfidazolam were randomized into 4 groups with 120 cases each. The concentration of midazolam in the culture meadium was 1 mg/L in group A, 10 mg/L in group B,and 20 mg/L in group C. Group D did not add midazolam as the control. Midazolam exposure was from 4.3-30 h post fertilization (hpf). Then confocal microscopy was used to examine the vascular development in vivo. Results Compared with group D, group A had no significant embryonic vascular development change, embryonic gastrulation stopped at phenotype in group C, and group B had serious defects in angiogenic sprouting of intersegmental vessels and posterior vasculature. Conclusion 10 mg/L midazolam exposure may inhibit the zebrafish embryonic vascular development.
