摘要
通过变性梯度凝胶电泳(DGGE),对天山乌鲁木齐河源1号冰川不同海拔表层雪及雪坑雪中真核微生物的多样性进行比较分析,并结合δ18O结果,探讨了天山乌鲁木齐河源1号冰川雪中真核微生物多样性分布及其时空特征.结果表明:天山乌鲁木齐河源1号冰川真核微生物分属于Viridiplantae,Fungi,Amoebozoa和Alveolata四大类,藻类为主要类群,且均为衣藻.序列比对结果显示,人类活动已明显影响到冰川中微生物的分布.同时,研究发现真核微生物多样性与海拔及雪样积累时间均呈负相关,与δ18O值呈正相关,表明温度是造成这种多样性分布时空变化的主要因素,暗示真核微生物同δ18O值一样,可作为气候环境变化的指标.
Surface snow samples of the different altitude and snow pits samples were collected from the Glacier No.1at therümqi River Head,Tianshan.Denaturing gradient gel electrophoresis(DGGE) were used to examine the diversity and its temporal-spatial characteristics of eukaryotic microorganisms with different altitude and depth by combine the δ18 O values.The results showed that the eukaryotic microorganisms belonged to four kingdoms:Viridiplantae,Fungi,Amoebozoa,and Alveolata.The algae was the dominant group,and all obtained sequences belonged to Chlamydomonadales.The diversity of eukaryotic microorganisms was negatively correlated with altitude and accumulated time,but positively correlated with the δ18 O values.Those results indicated that the temperature was the main factor for the temporalspatial change,and the diversity of eukaryotic microorganisms could be a index for the climate and environment change.
