体外培养成骨细胞的生长状态

Observation of the Osteoblasts Growth State in Vitro

摘要 目的:了解体外培养成骨细胞的生长状态,为其作为细胞模型研究骨替代材料提供依据。方法:将传代5次的兔成骨细胞,以1×105/mL、5×104/mL和1×104/mL的细胞密度接种于六孔板内,连续培养,倒置相差显微镜观察成骨细胞的生长状态。结果:各组成骨细胞在细胞贴壁、细胞形态、生长方式、增殖能力、钙化等方面有差异。结论:体外培养成骨细胞受接种时细胞密度的影响而呈现不同的生长状态。 To find out osteoblasts growth state in vitro for reasonable assessment of material biocompatibility and proper observation of biological behavior of osteoblasts,the fifth passage rabbit osteoblasts were inoculated in 6-well plates at the different cell density of 5×105/mL,1×105/mL,and 1×104/mL,and cultured in the DMEM medium with inverted microscope observation of cell growth state during the culture period.The observation found that the growth state of the osteoblasts in three cell density groups showed the differences in cell adherence,cell morphology,growth pattern,cell proliferation,and calcification.This suggests that there is a obvious effect of cell-inoculated density on the osteoblasts growth state in vitro.
