白假丝酵母菌分泌型天冬氨酸蛋白酶与重症婴幼儿龋的相关性 被引量:1

Relationship between secreted aspartyl proteinase of Candida albicans and severe early childhood caries

摘要 目的探讨白假丝酵母菌分泌型天冬氨酸蛋白酶(SAP)与重症婴幼儿龋(S-ECC)的相关性。方法以S-ECC患儿和无龋儿童口腔内的白假丝酵母菌临床分离菌株共40株为研究对象,分别采用YNB-BSA琼脂平板法及以牛血清白蛋白为底物配合四甲基偶氮唑盐法对比测定SAP蛋白酶活力,比较两组间SAP水平的差异;并分析SAP水平与25S rDNA基因分型的相关性。结果全部白假丝酵母菌菌株均表现为SAP蛋白酶阳性,且两种检测方法均显示S-ECC组的SAP蛋白酶水平(0.36±0.03、1.59±0.92)高于无龋组者(0.39±0.05、0.79±0.26),差异具有统计学意义(P=0.013、P<0.001);40株白假丝酵母菌25S rDNA基因型分为A、B、C三型,B型仅在S-ECC组出现,S-ECC组白假丝酵母菌A、B、C三型的SAP蛋白酶活力间差异无统计学意义(P=0.323、P=0.492)。结论白假丝酵母菌SAP表达水平升高可能与S-ECC的进展密切相关;但其酶活力高低与25SrDNA基因分型并无明显相关性。 Objective To investigate the relationship between secreted aspartyl proteinase (SAP) of Candida albicans and severe early childhood caries (S-ECC). Methods The activities of the SAPs from 40 strains of Candida albicans isolated from the oral cavity of children with S-ECC and without caries were evaluated contrastively by YNB-BSA agar plate method and BSA as substrate with MTT method. In addition,the association between the activities of SAPs and the genotypes of Candida albicans by 25S rDNA-PCR was analyzed. Results All strains secrete SAPs and the activities of the S-ECC group (0.36 ± 0.03,1.59 ± 0.92) were significantly higher than that of the caries-free group (0.39 ± 0.05,0.79 ± 0.26; P=0.013,P 0.001). The genotypes of Candida albicans were divided into groups A,B and C and the genotype B existed only in S-ECC group. The activities among genotypes A,B and C in S-ECC group were not statistically significant (P=0.323,P=0.492). Conclusions The SAPs of Candida albicans at higher level were closely associated with the progress of S-ECC. However,there was not a significant association between the activities of SAPs and the distribution of genotypes of Candida albicans strains.
