离子交换树脂分离纯化虫草素的工艺条件研究 被引量:13

Study on the Technical Parameters for Extracting and Purifying Cordycepin with Ion Exchange Resin

摘要 利用废弃蛹虫草培养基残基为原料提取虫草素,并以虫草素吸附量和解吸率为指标研究了虫草素在JK006阳离子交换树脂上的动态吸附参数,配合717阴离子交换树脂得到高纯度虫草素。结果表明:最佳吸附条件为:上柱液浓度为1.69mg/mL,上柱体积流量为3BV/h。最佳解吸条件为:氨水浓度0.25mol/L,洗脱体积流量2BV/h。经过717柱层析及重结晶后,虫草素的纯度可达到96.4%。该工艺操作简便,适合产业化生产。 Cordycepin was extracted and isolated from Cordyceps militaris solid medium.The adsorption and desorption rate of cordycepin were used as the index to study the optimum dynamic adsorption parameters of the JK006 cation exchange resin for cordycepin,combined with 717 anion exchange resin to obtain highly purified cordycepin.The resu1ts showed that the best conditions of dynamic adsorption are as follows:cordycepin concentration 1.69mg/mL and flow rate 3BV/h,and the best conditions of dynamic desorption:ammonia concentration 0.25mol/L,and flow rate 2BV/h.Purity of cordycepin reaches 96.4% through 717 column chromatography and recrystalization.The process is simple and suitable for industry production.
