逍遥散对慢性应激状态下大鼠海马神经细胞内糖皮质激素受体表达的影响 被引量:21

摘要 探讨逍遥散治疗血清对慢性应激大鼠状态下海马神经细胞内糖皮质激素受体蛋白表达的影响。方法:采用免疫印迹法检测慢性应激大鼠模型血清以及逍遥散治疗血清干预体外培养的海马神经细胞后,其胞内糖皮质激素受体(iGR)表达情况。结果:慢性应激大鼠模型血清与谷氨酸(Glu)共同作用于大鼠海马神经细胞对iGR表达下调的作用最明显,MK-801未能完全阻断iGR的表达下调,逍遥散治疗血清能够明显对抗慢性应激引起的iGR表达下调,但与空白组比较还存在差异,当与MK-801共同作用时,这种对抗iGR含量降低的作用倾向更加显著。结论:模型血清中可能存在Glu-NR以外途径的其他信号通路下调GR表达,逍遥散治疗血清对维持海马神经细胞iGR表达量的稳定起到一定作用,与MK-801可能协同性地促进慢性应激状态下海马负反馈调节功能的恢复。逍遥散可能通过包括Glu-NR-Ca2+-cAMP-iGR细胞信号传导通路在内的多种途径维持海马神经细胞iGR的稳态,以促进过度的应激反应尽快终止。 Objective:To study the effects of the serum of chronic stress rat model and the serum treated with Xiaoyao San on the expression of intracellular glucocorticoid receptor(iGR) in the cultured rat hippocampal nerve cells.Methods:The methods of Western blot and relative quantification was used to determine the protein expression of iGR in the cultured rat hippocampal nerve cells after being conducted by the serum of chronic stress rat model and the serum treated with Xiaoyao San.Results:The protein expression of iGR was decreased significantly when glutamate and serum of rat model with chronic stress were effected on the cultured rat hippocampal neurons;MK-801 had failed to downregulate the expression of iGR;Chronic stress-induced downregulation of iGR could be against significantly by pretreatment with the serum that treated with Xiaoyao San,though there still were differences between control group;The effects tend to be more significant when the cultured hippocampal neurons had been pretreated by MK-801 together with the serum treated with Xiaoyao San.Conclusion:Expression of NR2A and NR2B mRNA were closer to normal when MK-801 and serum of chronic stress treated with Xiaoyao San were used together.Conclusion:There may be some other signaling pathways other than that of Glu-NR that can downregulate the protein expression of iGR in the effects of serum of chronic stress rat model.Xiaoyao San had played a role of maintaining stability of the protein expression of iGR in hippocampal neurons when suffered with chronic stress,and it may work through a variety of signaling pathways including Glu-NR-Ca2+-cAMP-iGR to maintain the steady-state of hippocampal nerve cells.
