苯甲醛缩2-氨基嘧啶席夫碱铜配合物的合成及其与DNA相互作用

摘要 在非水溶剂中合成出一种新型席夫碱试剂(L):苯甲醛缩2-氨基嘧啶及其过渡金属铜配合物。由元素分析、络合滴定法、红外光谱、热重谱、质谱和摩尔电导值对配体和配合物的结构进行了表征,并推测出其组成为CuL2,采用荧光光谱和黏度分析方法初步研究了该配合物与小牛胸腺DNA之间的相互作用。表征结果表明:配合物为两分子配体以亚氨基和嘧啶环上的两个N原子与铜中心离子成键,配位数为4;与小牛胸腺DNA作用结果显示:配合物主要以插入方式与DNA相互作用,破坏了DNA的双螺旋结构。 In this paper,a novel Schiff base(L) derived from benzaldehyde and 2-aminopyrimidine and its copper complex have been synthesized in a nonaqueous solvent.Their molecular structures were characterized by elemental analysis,complexometric titration,IR,TG,MS and molar conductance measurements,and the formula of complex was confirmed to be CuL2.The interaction of the complex with calf thymus DNA was investigated by fluorescence spectroscopy and viscosity measurements.The results of structural characterization showed that the two ligands were bonded with copper central ion by two N atoms of imido group and pyrimidine ring,and the coordination number of the complex was four.The experiment relating to interaction with DNA indicated that the interaction of the complex with DNA was mainly of the intercalation mode,leading to damage of the double helix structure of DNA.
