玉竹不定芽诱导增殖培养影响因素的研究

Influencing Factors of Induction and Proliferation of Adventitious Buds of Polygonatum odoratum Druce

以初代培养获得的玉竹(Polygonatum odoratum Druce)带芽茎段为外植体,研究了不定芽诱导增殖的培养条件。结果表明:MS+6-BA 3 mg/L+NAA 0.1 mg/L和MS+6-BA 3 mg/L+IBA 0.1 mg/L能较好地直接诱导玉竹茎段形成不定芽;玉竹不定芽的增殖效应与基本培养基、糖浓度、pH值、光照时间和细胞分裂素均有较大关系,筛选出最佳的诱导和增殖培养条件为MS+NAA0.1mg/L+6-BA 3 mg/L+蔗糖30 g/L+琼脂7 g/L(pH6.0),光照时间16 h/d。

Using the initial culture stems with buds of Polygonatum odoratum Druce as explants,the culture conditions for induction and proliferation of adventitious buds were studied.The results showed that the best culture medium for direct induction of adventitious buds of Polygonatum odoratum Druce was MS＋6-BA 3 mg/L＋NAA 0.1 mg/L or MS＋6-BA 3 mg/L ＋IBA 0.1 mg/L.The basic culture medium,sucrose concentration,pH,photoperiod and cytokinin were significantly affected proliferation of adventitious buds of Polygonatum odoratum Druce.The best culture medium formula for induction and proliferation was MS ＋NAA 0.1 mg/L ＋6-BA 3 mg/L ＋sucrose 30 g/L ＋agar 7 g/L with pH 6.0 and photoperiod 16 h/d.
