P38MAPK与HO-1在急性肺损伤中的探讨

摘要 目的探讨丝裂原活化蛋白激酶P38(P38MAPK)与血红素加氧酶-1(HO-1)在急性肺损伤中的相互关系。方法健康雄性Wistar大鼠40只,随机分为5组(n=8)。对照组(NS组)、内毒素组(LPS组)、血晶素组(Hemin组)、锌原卟啉Ⅸ组(ZnPPⅨ组)、SB203580组(SB组)。于6h后用Western Blot和免疫组化法分别检测肺组织P38MAPK和HO-1蛋白表达,检测肺泡灌洗液(BALF)中中性粒细胞比、肺组织湿/干重、蛋白含量和动脉血气,并进行病理学观察。结果与NS组比较,LPS组、Hemin组、SB组、ZnPPⅨ组P38MAPK含量蛋白表达明显增高(P<0.05),HO-1强阳性表达(P<0.01或P<0.05),肺组织湿/干重明显增加(P<0.05),中性粒细胞比、蛋白含量显著增加(P<0.05或P<0.01),PaO2、PaCO2和HCO3ˉ较NS组显著下降(P<0.05);与LPS组比较,Hemin组、SB组P38MAPK蛋白表达显著降低(P<0.05),HO-1的表达明显升高(P<0.05),肺组织湿/干重、中性粒细胞比、蛋白含量明显减少(P<0.05),而ZnPPⅨ组恰好相反;Hemin组、SB组之间无显著差异(P>0.05)。ZnPPⅨ组肺组织损伤最重,LPS组次之,Hemin组和SB组最轻。结论 P38MAPK与HO-1在急性肺损伤中各自独立,相互抑制。
