细胞外基质修复兔阴茎白膜中iNOS和TGF-β1因子的表达及意义

Expression and Significance of iNOS and TGF-β1 in acellular matrix grafts for reconstructing tunica albuginea in rabbits

摘要 目的对SIS和BECM修复阴茎白膜缺损进行动态评价,观察不同时间点i NOS和TGF-β1的表达情况,旨在寻找理想的阴茎白膜修复重建材料。方法将60只新西兰兔分为对照组、SIS组和BECM组三组,每组20只,在阴茎背侧切除白膜10 mm×5 mm造成缺损,分别采用原位缝合、SIS或BECM修复。分别于术后2、6、12和24周处死动物术区取材:用免疫组化方法检测不同时间点炎性标志因子i NOS和促纤维化因子TGF-β1的表达情况。结果 iNOS和TGF-β1在2周时中等表达,6周时明显下降,2周和6周比较差异有统计学意义(P<0.05),此后表达趋于稳定,6周、12周和24周同组间相邻时间点或同一时间点不同组间i NOS和TGF-β1表达差异无统计学意义。结论 SIS和BECM在新西兰兔阴茎白膜的修复中显示了良好的组织相容性,未出现明显的炎症反应和纤维化,两种生物材料都是比较理想的阴茎白膜修复材料。 Objective To evaluate the morphological,immunological and functional response to small intestinal submucosa grafting and bladder extracellular matrix of the tunica albuginea to determine its potential as a grafting material for penile surgery.Methods Male New Zealand white rabbits were divided into three groups which were a sham procedure or tunical excision and grafting with small intestinal submucosa(SIS) and bladder extracellular matrix(BECM).There were 20 rabbits.The rabbits were different operations according to the study design,then were original point stitched,SIS or BECM repaired.the The animals were euthanized,and the penis was sectioned and histologically studied at intervals of 2,6,12,and 24 weeks.Protein expression of the inflammatory markers inducible nitric oxide synthase(iNOS) and transforming growth factor-β1(TGF-β1) were detected by immunohistochemical methods.Results Three groups showed middle expression of iNOS and TGF-β1 inducible at 2 week precipitously decreased to a normal appearing tunica albuginea at 6 and 12 and 24 week,there were no statistics significance difference in any groups.Conclusion Both SIS and BECM demonstrate tissue specific regeneration properties in the New Zealand white rabbits tunica albuginea.There are not remakable inflammation response or corporeal fibrosis.Both biological materials are safe,technically easy to use as penis repaired.
