昆明小鼠实验性自身免疫性脑脊髓炎模型的建立

摘要 用豚鼠脊髓匀浆液和不完全弗氏佐剂制备免疫原,联合腹腔注射百日咳毒素,建立昆明小鼠实验性自身免疫性脑脊髓炎动物模型。发病小鼠临床神经系统症状逐渐加重,脑组织经HE染色后,可见血管周围有大量炎性细胞浸润,呈袖套状改变;大脑白质可见胶质细胞增生形成胶质细胞结节以及神经细胞坏死等病理改变。正常小鼠中枢神经系统功能及病理检查均未见异常。此模型为进一步阐明多发性硬化的发病机制打下基础。 Chinese Kun Ming (KM) mice were immunized with both guinea pig spinal cord homogenate (GPSCH) in complete Freund' s adjuvant (CFA) and pertussis toxin (PTx) to establish an experimental autoimmune encephalomyelitis (EAE) model. EAE clinical symptoms gradually worsened with the development of this disease. Perivascular cuffs of inflammatory cell infiltration, formation of glial nodules and necrosis of neurons were observed in brain slices of EAE mice, which were stained with HE. By contrast, there were no signs of neural function disorder and parenchymal pathology in mice from the adjuvant group. This EAE model provided in this paper will be helpful to illuminate the pathogenesis of multiple sclerosis (MS).
