摘要
目的探讨甲醛致小鼠卵巢细胞DNA损伤效应及对Bcl-2和Bax表达的影响与其相关作用的机制。方法采用单细胞凝胶电泳方法,检测甲醛染毒(剂量分别为0.3、1.2、4.8 mg/kg)后小鼠卵巢细胞DNA损伤情况,采用免疫组织化学法检测Bcl-2、Bax的表达。结果甲醛致卵巢组织结构损伤,染毒浓度越高,病理损害越明显;卵巢细胞Bcl-2下调,Bax表达上调,并有量-效关系,甲醛染毒各组Bcl-2和Bax表达的相对灰度值较对照组均有差异(P<0.01);试剂量范围内,小鼠卵巢细胞彗星率随甲醛染毒剂量增加而增高(P<0.05),其彗星尾长随甲醛染毒剂量增加而缩短,尤其高甲醛染毒组更短(P<0.01)。结论甲醛能致卵巢细胞DNA损伤效应,损伤小鼠卵巢的结构,使卵巢细胞凋亡增加,Bcl-2表达下调、Bax表达上调;Bcl-2和Bax表达的变化可能是诱导凋亡的机制。
Objective To study the effect of formaldehyde on DNA damage and expression of Bcl-2 and Bax in ovary cells of mice.Methods The effect on DNA damage in the ovary cells after peritoneal injection of formaldehyde(0.3,1.2,4.8 mg/kg) were measured by using the single cell gel electrophoresis technique(comet assay)tests,the expression of Bcl-2 and Bax were tested by immunohistochemical method.Results In treatment groups of formaldehyde,the ovarian morphosis was damaged,the expression of Bcl-2 was degression and that of Bax was enhancement,and there were a significant dose-effect relationship and significance of difference(P〈0.01)compared with the control group.Within tested doses,the mumber of comet cells in the ovary cells was significandy increased as formaldehyde exposue dose increased(P0.05).The length of DNA migration was significantly shortened with formaldehyde increasing,especially high formaldehyde dose(P〈0.01).Conclusion Formaldehyde could promote the response of DNA damage of ovary cells,damage ovarian morphosis,cause apoptosis to raise,depress the expression of Bcl-2 and enhance the expression of Bax in mouse ovarian.The mechanism of apoptosis may be related to the abnormal expression of Bcl-2 and Bax in mouse ovarian.
