Establishment of experimental animal model of osteoporosis with rat by ovariectomy

摘要 目的建立大鼠卵巢去势骨质疏松模型模拟妇女绝经后骨结构变化。方法 20只3月龄雌性SD大鼠随机均分为2组:假手术组(Sham,n=10)、卵巢去势组(OVX,n=10),分别进行假手术和卵巢去势术,12周后活体测量腰椎、胫骨骨密度,处死动物后取胫骨行扫描电镜观察,取子宫称重。结果腰椎、胫骨骨密度检测显示OVX组出现系统性骨密度下降,Sham组为(302.50±25.92)g/cm3,(205.90±15.07)g/cm3;OVX组为(264.10±20.10)g/cm3,(163.20±11.15)g/cm3,Sham组显著高于OVX组(P<0.05);电镜下骨形态学表现也支持骨密度测量结果;Sham组大鼠体质量低于OVX组,而子宫质量和子宫系数均高于OVX组。结论大鼠卵巢去势骨质疏松模型建立成功。 Objective To imitate bone structure of postmenopausal woman by establishing experimental animal model of osteoporosis with rats.Methods Twenty female SD rats were randomly divided into 2 groups: sham-operated group(n=10)and ovariectomized group(n=10) which were submitted to sham surgery or ovariectomy according to their designations.After 12 weeks densitometry of lumbar and tibiae was performed by dual-energy X-ray absorptiometry and tibiae was subjected to observation by SEM.Uterus was obtained and weighted.Results Densitometric analysis of the lumbar and tibiae confirmed a systemic impairment of animals,disclosing lower values(P0.05) of bone mineral density for OVX[(264.10±20.10)g/cm^3,(163.20±11.15)g/cm^3]in relation to the sham group[(302.50±25.92)g/cm^3,(205.90±15.07)g/cm^3].The observation under SEM supports results of bone density measurement.Weight of sham group was lower than OVX group,Uterus weight and coefficient of uterus were all higher than OVX group.Conclusion It was successful for establishment of experimental animal model of osteoporosis with rat by ovariectomy.
