摘要
目的调查山东省济宁地区人群16个Y-STR基因座及其单倍型的遗传多态性,探讨其法学应用价值,为法医学应用提供基础数据。方法应用AmpFlSTR Yfiler试剂盒,对济宁地区197名汉族无关男性个体进行16个Y-STR基因座的复合扩增,用ABI3130型基因分析仪对扩增产物进行检测,统计其群体遗传学参数。结果前15个基因座分别检出6、5、6、6、8、6、6、3、7、8、6、4、3、4、6个等位基因。DYS385 a检出10个等位基因,DYS385 b有165例表达,检出14个等位基因,DYS385 a/b基因座检出47种单倍型。基因多样性(GD)值最低为0.3867(DYS438),最高为0.9717(DYS385 a/b)。在165例全表达对象中,观察到16个Y-STR基因座共同构成的单倍型160种,其中159种单倍型只出现1次,5种出现2次,1种出现3次,累计GD值为0.9994。结论 16个Y-STR基因座具有较强的个体识别能力,可应用于法庭科学中的个体识别与亲权鉴定。
Objective To investigate the genetic polymorphism of 16 Y-chromosome STR loci and haplotype in Han population in Jining area,and to evaluate its forensic application values,which will provide the base date for forensic application.Methods DYS456,DYS390,DYS389Ⅰ,DYS389Ⅱ,DYS458,DYS19,DYS393,DYS391,DYS439,DYS635,DYS392,GATA-H4,DYS437,DYS438,DYS448 and DYS385 loci were amplified with AmpFlSTR Yfiler kit and analyzed by ABI 3130 genetic analyzer.197 unrelated male individuals of the population in Jining area were investigated to determine the distributions of allele frequencies and haplotype.Results 6,5,6,6,8,6,6,3,7,8,6,4,3,4,6 alleles were found across the Y-STR loci.10 alleles were detected across DYS385a,14 alleles across DYS385b,among those 165 expressed object,and 47haplotypes across DYS385a/b locus.The minimum GD value was 0.3867(DYS438),and the maximum was 0.9717(DYS385a/b).160 haplotypes were found in 165 comp-letely expressed objects,of which 159 were unique,5 haplotypes were shared by 2 individuals,1 haplotype was found in 3 individuals,and the observed haplotypes diversity value was 0.9994.Conclusion The 16 Y-STR loci are highly polymorphicand suitable for forensic individual identification and paternity testing.
