摘要
cry1Ia1基因编码的是苏云金芽孢杆菌(Bacillus thuringiensis,简称Bt)的杀虫晶体蛋白(ICPs),产物对鳞翅目(Lepidoptera)和鞘翅目(Coleoptera)昆虫具有毒害作用,可防治大豆食心虫等害虫。利用农杆菌介导的方法将cry1Ia1基因转入大豆栽培品种黑农35中,并获得了5株转cry1Ia1基因的株系。PCR、Southern blotting和RT-PCR检测结果表明cry1Ia1基因已经整合到大豆基因组中并稳定表达。通过人工接种的方法对转基因植株抗大豆食心虫的水平进行了初步的评价,结果表明其中的2株对大豆食心虫具有高度的抗性,2株具有中等程度的抗性,1株与对照无明显差异。cry1Ia1基因首次成功导入大豆栽培品种黑农35中并获得抗大豆食心虫的转基因植株。
The cry1Ial is derived from Bacillus thuringiensis, encoding insecticidal control proteins (ICPs) which is toxic to Lepi- doptera and Coleoptera insects, and can be used against soybean pod borer. The cry1Ial was successfully transferred into soybean cuhivar Heinong 35 by Agrobacterium-mediated transformation, and five soybean transgenic lines were acquired. The PCR, Southern blotting and RT-PCR results indicated that the cry1Ial was integrated into soybean genome and expressed by its promoter. The resis- tance to soybean pod borer was estimated by artificially introducing soybean pod borer, and two lines of transgenic soybean plants showed high resistance to soybean pod borer and another two lines with middle resistance, and one line with no difference tO that of the control. The cry1Ial was first introduced into soybean cuhivar Heinong 35 and transgenic soybean plants against soybean pod borer was obtained.
