摘要
目的通过对脓毒症模型的治疗过程,观察人类杀菌蛋白(BP)的治疗作用,并与抗LPS单克隆抗体1A比较。方法脓毒症模型采用NH1雄性小鼠腹腔注射O111B4大肠杆菌悬液。给药分三组,BP01mg,1A10mg,生理盐水(NS)05ml。结果各组生存率无显著性差异(P>005)。第二次攻击后3小时外周血白细胞计数,NS组稍高于其他两组,分别为196±57×106/ml、155±38×106/ml和152±30×106/ml,差异未达到显著性界值。血浆LPS水平:BP和lA组明显低于NS组(P<005)。血浆细胞因子活性:BP组对细胞因子的抑制作用较强,分别在不同时间点对IL-1、IL-6和TNF的数据低于NS组,有显著性差异(P<005);lA只在一个时间对IL-6有抑制作用(P<005)。结论抗LPS单抗和BP对LPS均具有阻断作用,但是,BP的作用更强。两者均不能明显改善动物的生存率,应作为辅助治疗。
Objective Therapeutic
effects were tested in the treatment of septic mice with bactericidal protein (BP), compared with
anti-LPS monoclonal antibody 1A. Methods NH1 male mice were challenged with
intraperitonial(IP)bolus injection of E.Coli O111 B4 suspension .BP at 0.1mg and 1A at 1.0mg
were administrated IP,controlled by the same injected volume of normal saline (NS).Survival
and white blood cell of peripheral blood were investigated .Cytokines activities of septic mice
were measured respectively by enhancing the mitogenic response of mouse thymocytes to Con
A for IL-1,IL-6-dependent murine hybridoma cell line 7TD1 for IL-6, and L929 cytolytic assay for
TNF.Blood LPS level was measured by a quantitative chromogenic limulusamebocyte lysate
test. Results There was lower blood LPS level in both lA and BP groups than in NS control( P
<0.05). Compared with 1A,BP has much stronger endotoxin-neutralizing ability,which show
lower blood level of IL-1,IL-6 and TNF in BP group than in lA group.But there were no significant
differences in survival percentage and WBC (P >0.05).Conclusions The results suggested that
BP might be a more powerful anti-inflammatory agent than anti-LPS antibody in the treatment of
patient with bacteria infection . [
