三氧化二砷对肝癌HepG2细胞株RhoC、CD44v6及Ezrin基因表达的影响 被引量:4

Effects of arsenic trioxide on the expressions of RhoC, CD44v6 and Ezrin in human hepatocellular carcinoma cells in vitro

摘要 目的观察三氧化二砷(As2O3)对肝癌HepG2细胞中RhoC、CD44v6和Ezrin表达的影响,探讨As2O3抑制肝癌细胞侵袭、转移的机制。方法采用RT—PCR法检测As2O3作用后RhoC、CD44v6及EzrinmRNA表达水平的变化。以Western blot方法检测RhoC基因蛋白表达。结果0.25mg/LAs2O3作用4、6、8d后HepG2细胞RhoC mRNA及蛋白、CD44v6mRNA表达水平较药物作用前均有不同程度降低(p〈0.05),EzrinmRNA表达水平均无明显变化(P〉0.05)。结论As2O3,并非通过单一靶点发挥抗肝癌侵袭转移作用,可能与下调RhoC及CD44v6基因的表达有关。 Objective To investigate the effects of arsenic trioxide on the expressions of RhoC, CD44v6 and Ezrin in human hepatocellular carcinoma cells (HepG2) in vitro and the mechanism by which arsenic trioxide inhibits the invasion and metastasis of hepatocellular carcinoma cells. Methods The mRNA expressions of RhoC, CD44v6 and Ezrin were detected by RT - PCR, and the protein expres- sion of RhoC was examined by Western blotting in HepG2 cells. Results The mRNA expressions of RhoC and CD44v6 and the RhoC protein expression were significantly decreased in HepG2 ce|ls after treatment with 0.25 mg/L arsenic trioxide for 4,6 and 8 days ( P 〈 0.05 for all ). No significant change was found in the expression level of Ezrin mRNA in HepG2 cells treated by arsenic trioxide for 4,6 and 8 days when compared with that before the treatment ( P 〉 0.05 ). Conclusion Arsenic trioxide may reduce the metastatic potential of HepG2 ceils by down - regulating the RhoC and CD44v6 gene expressions.
