摘要
我们应用凝胶过滤及离子交换柱层板法对地鼠肾细胞狂犬病疫苗进行纯化,结果表明,凝胶柱层析法即可使该疫苗效价达到4.8IU/ml,小牛血清残留置小于50ng/ml,符合《中国生物制品规程》要求。并经离子交换柱层板后,效力降低至1.9IU/ml,病毒损失较大。故只采用凝胶过滤柱层析法做为该苗的纯化工艺,并为大规模生产提供依据。
In this paper,gel filtration chromatography (GFC) and ionexchange chromatography (IEC) were used to purify the rabies vaccine developed from Hamster kidney cells.The results indicated that to titer could reach to 4.8 IU/ml with GFC method a higher virus titer,in addition,fetal calf serum residue was less than 50ng/ml,which is in conformity with the requests of “ the Procedures of China Biological Products ”.However,using IEC method again after GFC resulted in a great deal of decrease of vaccine virus titer,which was 1.9IU/ml.The lost amount of the rabies viruses was great.Therefore,only the GFC used in the paper to purify the rabies vaccine developed from Hamster kidney cells is an effective,and with wiich a practicable method.Furthermore,this paper also provides a technique basis for the manufacture of vaccines on a large scale.
