三羟基苯甲酸纯化物及其化合物对Jurkat细胞周期进程及凋亡的影响 被引量:4

摘要 目的:研究不同浓度菱角三羟基苯甲酸(TBA)纯化物及其化合物对人T淋巴细胞白血病Jurkat细胞周期进程及凋亡的影响,并探讨2种不同来源TBA对肿瘤生长抑制的可能机制。方法:2种不同来源TBA分别作用于对数生长期的Jurkat细胞,每种TBA分为5组(0、3.125、6.250、12.500和25.000mg·L-1),作用于Jurkat细胞30h后,经流式细胞仪分别检测不同浓度菱角TBA纯化物及其化合物处理的Jurkat细胞周期各时相及其凋亡率变化。结果:Jurkat细胞分别经过不同浓度菱角TBA纯化物处理后,细胞凋亡率明显高于0mg·L-1组(P<0.05或P<0.01);与对照组比较,3.125mg·L-1组变化无差异;6.250、12.500和25.000mg·L-1组变化有差异(P<0.05或P<0.01)。Jurkat细胞分别经过不同浓度TBA化合物处理后,与对照组比较,细胞凋亡率在3.125mg·L-1组时即存在差异(P<0.05),随其剂量增加而增加(P<0.05或P<0.01),各用药组间变化无差异。经TBA纯化物作用后,G1期细胞百分数明显高于对照组(P<0.01),并随TBA剂量增加而增加,在6.250~25.000mg·L-1浓度下S期细胞百分数明显低于对照组(P<0.05或P<0.01),G2期无显著变化。经TBA化合物作用后在6.250~25.000mg·L-1浓度下G1期百分数明显高于对照组(P<0.05或P<0.01),S期细胞百分数明显低于对照组(P<0.05),G2期无显著变化。各用药组间变化无差异。结论:2种不同来源TBA均能够诱导细胞凋亡;均能够阻止人T淋巴细胞白血病细胞的G1期细胞向S期进程,其抑制作用可能通过G0/G1期阻滞引起。 Objective To study the effects of purified product of trihydroxybenzoic acid(TBA)from water caltrop and its compound at different concentrations on cell cycle progression and apoptosis of Jurkat cells in vitro and explore their possible mechanism of inhibiting tumor growth.Methods The Jurkat cells at logarithmic growth phase were treated with purified product of TBA and its compound in vitro.Each TBA was divided into 5groups (0,3.125,6.250,12.500and 25.000mg·L-1).The changes of cell cycle progression and apoptotic rate of Jurkat cells 30hafter treated with TBA were measured with flow cytometry.Results The apoptotic rates of the Jurkat cells treated with purified product TBA with doses of 3.125and 25.000mg·L-1 were increased significantly as compared with those in control group (P〈0.05or P〈0.01);the apoptotic rates of Jurkat cells treated with TAB compound with different doses were increased with the increase of doses(P〈0.05or P〈0.01).The percentages of G1phase cells treated with purified product TBA with different doses were increased significantly with the increase of doses as compared with those in control group (P〈0.01),the percentages of S phase cells were decreased significantly in 6.250and 25.000mg·L-1 groups(P〈0.05or P〈0.01),and the percentages of G2phase cells did not change significantly.The percentages of G1phase cells treated with TAB compound with 6.250and 25.000mg·L-1 were significantly higher than those in control group (P〈0.05or P〈0.01),the percentages of S phase cells were decreased significantly(P〈0.05),and the percentages of G2phase cells did not change significantly.Conclusion Two different TBA could all induce Jurkat cell apoptosis and G1phase arrest. Their inhibitory effects on tumor growth could be due to apoptosis caused by G1arrest.
